Treatments of depressive disorders have always been the focus from both clinical physicians and basic researchers. Although the therapeutic drugs for depression have evolved from the tricyclic depressants to SSRIs with the drug toxicity largely improved, there are still many problems awaiting for addressing regarding the current antidepressants. The authors in this article have summarized the evidence on nuclear receptors (NRs),including glucocorticoid receptor, mineralocorticoid receptor, estrogen receptor, aryl hydrocarbon receptor, and peroxisome proliferator-activated receptor, in modulating the inflammasome activity during development of depression from an endocrine perspective. This could enhance our understanding of how depression develops and might promote novel-drug developments.
Since drug safety without compromising efficacy is maximally pursued in clinic, how these nuclear receptors modulators, such as hormones and metabolites, would efficiently penetrate the brain-blood barrier remains to be elucidated from both preclinical and population-based cohort studies. It would be interesting to see more studies concerning the therapeutic efficacy and safety of nuclear receptor modulators in treatment of major depression.
